However, TDF as a Biopharmaceutics Drug Classification (BSC) class Ⅱ active compound, is poorly water soluble and highly degraded by CYP3A4 in liver, which can significantly affect the bioavailability and efficacy of the drug [[3], [4], [5]]. Various pharmaceutical technologies have been developed for increasing the solubility and bioavailability of chemical drugs [[6], [7], [8], [9], [10], [11], [12], [13], [14]]. Orodispersible film (ODF) presents as a thin film, which is simply placed on the patient's tongue or mucosal tissue, instantly wet by saliva; then it rapidly disintegrates and dissolves to release the medication for mucosal or gastrointestinal absorption [[30], [31], [32], [33], [34], [35], [36]]. More importantly, ODFs offer fast, accurate dosing in a safe, efficacious format that is convenient and portable, without the need for water or measuring devices [32,33]. Generally, ODFs are consist of drug, water soluble polymer, plasticizers, fillers, colors and flavors, etc. Especially, the polymer type and filler can significantly affect the disintegration and dissolution of ODFs [33,38]. As an oral fast dissolving drug delivery system, ODFs was developed rapidly recently and is being commonly used because of convenient usage and rapid onset of action with increased bioavailability by reducing or avoiding hepatic first pass effect and stability [39]. In order to overcome the problems of dissolution and bioavailability of insoluble drugs, more and more powders containing micronized drug particles are used in pharmaceutical preparations [[45], [46], [47]].

Therefore, this study is the first attempt to combine tadalafil micronization technology with ODF technology to improve oral bioavailability of insoluble drugs and improve patient compliance. Firstly, a novel TDF-loaded ODF with enhanced dissolution rate was prepared by using a solvent casting method with polyvinyl alcohol 1788 (PVA1788) as a film-forming material, sodium alginate (ALG-Na) as a disintegrant, polyethylene glycol 400 (PEG400) as a plasticizer, and sodium lauryl sulfate (SLS) as a surfactant.
